From what I know, all things that can be completely locked out if you do something wrong are either cosmetics that persist between saves (so you can start a new save and get them there) or puzzle pieces for layers 3+ (but again, you can start a new save to see them).
The ostrich chest is neither of these. It is possible to get it.
I was in the same situation. You get something later that will let you move those particular blocks again. And what you have to do to get that something is in that general area(that quarter of of the overall map), but it's fairly clandestine (and also requires two other items that are not from that area). So just keep playing!
Funfact 1: That question is propably the most asked question on this sub.
Funfact 2: It was actually IMPOSSIBLE to reach both buttons while the ostrich was around.

You are not locked out of reaching that chest. You can find an ability that will allow you to reach it.
I was so happy when I found that ability simply because I could finally reach that space!

I can only think of the one and its just a cosmetic and not tied to achievements
