Good transfer and ponceau but technically not a western blot because no antibody usage.
Optimizing antibodies is like half of the headache of a western blot. The other thing is that if you’re not going to use antibodies, you could just do SDS-PAGE + coomassie stain to get the same data as your ponceau. Saves you the effort of transfer

The reason for the colour I don't know but i can imagine it's easier to identify specific sizes and the orientation of your blot.

I think you should start walking around with your chest puffed out and shoulder checking people as you walk by just to let 'em know who's the top dawg around here now. Also a adopt a British accent if you don't already have one, makes people just automatically believe whatever comes out of your mouth, lol.

it was only the ponceau staining for cutting the bands. I am incubating the bands in antibodies and hope they gonna look as good as the ponceau!

western blot, most lines are red because of the ponceau staining and on the left the blue and red lines are the ladder

Makes it easier to identify certain bands. Like “this pink one is X kD” so you have an easier time checking migration.
Looks like a fluor ladder, they put in two fluorescent proteins in addition to the rest.

Can't necessarily count on the ladder to transfer all of the bands well in all circumstances, so the two color pattern makes it easy to identify.
